Time-Gated Fluorescence Correlation Spectroscopy for Improved Concentration Determinations

چکیده

Fluorescence Correlation Spectroscopy (FCS) is a very popular method to determine the diffusion coefficient of molecules in solution. However, the information contained in a FCS curve can be further exploited: As the mean number of molecules in the confocal detection volume Veff is inversely proportional to the amplitude of the correlation function at very short correlation times G (τ ≈ 0), FCS also allows to measure the concentration of the fluorescing molecules in the sample [1,2]. This is described by equation 1, if excitation into triplet states of the fluorophore and the background signal can be neglected.
